Why allergen detection by ELISA is insufficient to protect allergic consumers

چکیده
Background In 2010, a multi-allergen detection method based on mass spectrometric (LC-MS/MS) analysis was developed as described in JCA1218. This method was able to demonstrate that seven allergens could be detected in a single analysis. Later, it could be shown that the same method is capable to detect allergens in processed materials while conventional methods like ELSIA and PCR failed in the same matrix, even when amounts as high as 1000mg/kg were present. This was reported in JAOAC 94/4. In mid-2012 a case of a finished product was reported, which caused allergic reactions in children. The product was analysed by ELISA and LC-MS/MS. The ELISA failed to detect the allergens while mass spectrometric analysis clearly demonstrated the presence. This is the first case where evidence suggests that conventional methods like ELISA and PCR are not sensitive enough in some processed materials to protect allergic individuals, while LCMS/MS is still able to detect these allergens.

Quantification of Pla or 3, a Platanus orientalis Allergen, Grown under Different Environmental Conditions, by Sandwich ELISA
Background: Platanus species are widely cultured around the world and considered an important cause of allergic reactions. In the present study, we developed a sandwich ELISA to quantify Pla or 3 allergen in P. orientalis pollen extracts grown near high-traffic roads and compared it to pollen extracts collected from rural areas as control. Methods: Pollen samples were collected from three po...

The efficacy of allergen immunotherapy for the treatment of allergic rhinoconjunctivitis with or without seasonal bronchial asthma and anaphylaxis caused by the sting of the hymenoptera class of insects has been clearly demonstrated in numerous well-designed, placebo-controlled trials. Immunotherapy whether by subcutaneous injection of allergen extract or by oral/sublingual routes modifies peri...

متن کاملDetection of Allergen Markers in Food: Analytical Methods
Food allergens are proteins that are well tolerated by most, but can cause severe reactions in sensitive individuals. Since there is no cure for food allergy, strict adherence to an allergen-free diet is the only safe choice currently available for allergic consumers. Accurate food labeling can help consumers avoid foods containing an allergenic ingredient. Regulatory agencies have mandated the...
